Ischemic stroke (IS) is a serious neurological condition with complex pathological mechanisms. Tetrahydropiperine (THP), a natural alkaloid, has demonstrated protective effects against various diseases, including seizures and pain. This study investigates the potential neuroprotective effects of THP in ischemic stroke and explores its underlying mechanisms.
Using network pharmacology and molecular docking techniques, we identified potential target proteins of THP for intervention in IS. Network pharmacology analysis suggested that THP's effects on IS are particularly prominent within the PI3K/Akt signaling pathway. Molecular docking results showed that THP has a strong binding affinity for proteins involved in autophagy and the PI3K/Akt/mTOR pathway.
Our results indicated that THP significantly improved behavioral outcomes, reduced cerebral infarction size, alleviated histopathological damage from ischemia, enhanced neuronal survival, and reduced ultrastructural neuronal damage (P < 0.05). Additionally, THP promoted the survival of PC-12 cells exposed to oxygen-glucose deprivation (OGD) and improved their morphological integrity (P < 0.05). Furthermore, THP increased the levels of P62, LC3-I, PI3K, P-Akt/Akt, and P-mTOR/mTOR proteins, while decreasing the levels of Atg7 and Beclin1 proteins.
