Retinol, widely used for its anti-wrinkle properties in cosmetics, faces stability issues due to oxidation when exposed to air. To address this challenge, retinol-loaded lipid nanocarriers were synthesized using the vacuum emulsification method. This approach improves retinol's stability, protects it from oxidative degradation, and enhances its skin penetration efficiency.
Synthesis of Retinol-Loaded Lipid Nanoparticles
Lipid nanocarriers were prepared using the vacuum emulsification method. Initially, solid lipids, including glyceryl behenate, glyceryl stearate, glyceryl distearate, cetyl palmitate, and myristyl myristate, were melted at 75°C with hydrogenated lecithin to promote efficient solubilization while stirring gently. A separate mixture of caprylic/capric triglyceride, BHT, and retinol was heated at 50°C with magnetic stirring until fully dissolved.
Simultaneously, three surfactants-Tween-60, poloxamer 188, and sodium stearoyl glutamate-were added to distilled water, which was heated to above 80°C and stirred at approximately 500 rpm. Once both lipid mixtures were fully melted, the retinol-containing liquid lipid mixture was added to the solid lipid mixture.
Next, the hot aqueous phase, containing surfactants and additives, was combined with the lipid solution in a vacuum homogenizer. The mixture was emulsified for 5 minutes under vigorous stirring. The final lipid nanoparticle dispersion was cooled to 25°C and stored at 4°C until further use.
