The Effect of Red Raspberry Ketone (RRK) on Serum Liver Injury Markers
After liver injury, AST and ALT are released from the liver, reflecting the degree of liver damage. As shown in Figure A-B, the AST level in the MC group rats was significantly higher than that in the NC group (P < 0.01), while the AST levels in the PC group and RRK group were lower than that in the MC group (P < 0.01). In addition, the ALT level in the MC group also showed a significant improvement compared to the normal group (P < 0.01), while the ALT levels in the PC group and RRK treatment group were very significantly decreased compared to the MC group (P < 0.01). Therefore, RRK can reduce the serum levels of ALT and AST, and it is speculated that RRK may repair the necrotic hepatocytes to a certain extent and may reduce the permeability of the cell membrane. Overall, RRK can inhibit the further development of non-alcoholic fatty liver disease (NAFLD) and has certain clinical preventive significance.

The Effect of RK on Melanogenesis
Treatment of B16 cells with RK and direct monitoring of melanin content in the treated cells by Fontana-Masson staining showed the results as depicted in Figure A. IBMX (a cAMP-elevating agent) was used to stimulate melanogenesis in B16 cells. Two melanogenesis inhibitors, arbutin and deoxyarbutin, were used as positive controls in B16 cells. After IBMX treatment, the melanin content in the cells increased, and both arbutin and deoxyarbutin treatments reduced the increase in melanin content. Compared to IBMX-stimulated cells, all RK treatments also significantly decreased the melanin content in the treated cells. The melanin content in the treated cells was also quantified by a spectrophotometric method that detects melanin in cell lysates at 490 nm after NaOH solubilization (Figure B). The obtained curves were similar to those from the Fontana-Masson staining, clearly showing the dose-dependent inhibition of melanogenesis by RK. Potential cytotoxic effects of RK on B16 cells were tested by treating cells with different concentrations of RK and measuring cell viability by the MTT method. As shown in Figure C, although RK strongly inhibited melanogenesis in murine B16 melanoma cells, it did not exhibit any cytotoxicity.
