Neryl butyrate is a component of the volatile oil extracted from aromatic plants. Structurally similar aliphatic organic compounds with properties close to neryl butyrate have shown vasodilatory effects in the rat aorta. To evaluate whether neryl butyrate also has a relaxant effect, this study tested its influence on isolated rat aortic preparations.
Unlike the similar compounds, neryl butyrate did not exhibit any relaxant effect in pre-contracted aortic rings with phenylephrine, but instead induced contraction when applied to the resting aortic rings. The contractile effect was enhanced in the endothelium-denuded aortic rings. Treatment of the intact preparations with the nitric oxide synthase inhibitor L-NAME or the guanylate cyclase inhibitor ODQ also enhanced the contractile response to neryl butyrate. This phenomenon was not observed in the presence of the cyclooxygenase inhibitor ndomethacin. The contractile response was reduced in the presence of the L-type Ca2+ channel blocker verapamil or when Ca2+ was removed from the extracellular solution. The α-adrenoceptor antagonists prazosin and yohimbine reversed the contraction induced by neryl butyrate, but the thromboxane-prostaglandin receptor antagonist seratrodast did not. The α1A-selective antagonist RS-17053 antagonized the contraction induced by neryl butyrate. Inhibition of phospholipase C or Rho-associated kinase using U-73122 or Y-27632, respectively, reduced the contraction evoked by neryl butyrate. Intravenous injection of neryl butyrate in conscious rats caused hypotension and bradycardia. A reduction in heart rate was also observed in isolated right atrial preparations. In summary, the contractile effect of neryl butyrate is inhibited by α-adrenoceptor antagonists, indicating the involvement of α-adrenoceptors in the mechanism of action. In vivo, neryl butyrate induces hypotension, suggesting systemic effects beyond vascular contraction.
