To achieve nanoscale particle formation, the emulsion was ultrasonicated at 60% amplitude using a probe-type ultrasonic homogenizer for varying durations (5-10 minutes). After ultrasonication, the dispersion was rapidly cooled in an ice bath (2-3°C) for 5 minutes to induce particle solidification. Different formulation parameters, including lipid-to-oil ratios (1:1, 1.5:1, or 3:1 w/w), total lipid concentrations (2-4% w/v), and P407 concentrations (1-2% w/v), were systematically optimized to obtain stable dispersions.
For solid-state characterization, MJ-NLC dispersions were frozen at -80°C for 24 hours and lyophilized under vacuum (0.4 bar) for 24 hours. The optimized MJ-NLCs were further used as a lipid matrix for ivermectin co-loading, demonstrating their potential as an advanced nanocarrier system. This study underscores the precise formulation strategies required to develop stable and effective lipid-based drug delivery platforms.
