Isopropyl sorbate was employed as an essential component in the synthesis of injectable methacrylate gelatin (GelMA) microspheres (GM) functionalized with dopamine and hepatocyte growth factor (HGF) for tendon regeneration. The GM were fabricated using a microfluidic device, where isopropyl sorbate (5% w/w) served as part of the oil phase in the emulsification process. The oil phase, consisting of isopropyl sorbate and Span80, was mixed with the aqueous phase containing GelMA and photoinitiator in phosphate-buffered saline (PBS). The coaxial nozzle setup allowed for controlled injection rates, and the resulting emulsions formed microspheres upon UV cross-linking.
After cross-linking, the microspheres were thoroughly washed with deionized water and acetone to remove residual isopropyl sorbate and Span80, ensuring the purification of the final product. The functionalized GM were then immersed in a HDC solution, followed by the adsorption of HGF to form the GM@HDC@HGF composite. This formulation demonstrates the pivotal role of isopropyl sorbate in facilitating the formation of stable and functionalized GelMA microspheres. The resulting GM@HDC@HGF microspheres hold promise in modulating the tendon microenvironment and preventing tendon degeneration in tendinopathy therapies.
