Dietary isoprenoids, including β-ionone, represent a class of promising chemopreventive and chemotherapeutic agents. Particularly, β-ionone has garnered significant attention due to its selective cytotoxicity against tumor cells and its anti-metastatic and apoptosis-inducing properties both in vitro and in vivo.
Gastric Cancer
The viability and apoptosis of gastric adenocarcinoma cells (SGC-7901 cell line) were evaluated after treatment with gradually increasing concentrations (25, 50, 100, and 200 μM) of β-ionone for 24 and 48 hours. The observed growth inhibition of SGC-7901 cells by β-ionone was 12.04%, 30.59%, 78.25%, and 94.15%, respectively.
Breast Cancer
To investigate the anti-proliferative effects of β-ionone on breast cancer cells, human breast cancer cells (MCF-7) were treated with different concentrations of β-ionone (25, 50, 100, and 200 μM), and cell growth inhibition, cell mitosis, colony formation, and DNA synthesis were evaluated. β-ionone inhibited the proliferation of MCF-7 cells in a dose-dependent manner, with an IC50 value of 104 μM.
Prostate Cancer
The cytotoxic activity of β-ionone was evaluated against DU145 and LNCaP (human prostate cancer cells) and PC-3 (prostate adenocarcinoma cells). The results showed that β-ionone inhibited the proliferation of DU145 (IC50 = 210 μM), LNCaP (IC50 = 130 μM), and PC-3 cells (IC50 = 130 μM) in a concentration-dependent manner.
Liver Cancer
The anti-metastatic effects of β-ionone and its underlying mechanisms were evaluated in highly metastatic human liver cancer SK-Hep-1 cells. The data showed that incubation of SK-Hep-1 cells with β-ionone (1-50 μM) for 24 and 48 hours significantly inhibited cell invasion, migration, and adhesion.

Cyclooxygenase-2 (COX-2) as a potential target for cancer chemoprevention plays a key role in tumor cell proliferation and carcinogenesis. Therefore, the purpose of this study was to determine the activity of β-ionone (BI) in inhibiting COX-2 in breast cancer and its relevance to cancer cell models.
β-Ionone Reduces COX-2 Activity
To explore the possible mechanism by which BI inhibits cancer cell proliferation, the expression of COX-2 was measured in cell models. PMA significantly induced high expression of COX-2 in SGC-7901 cells. After 6 hours of treatment with PMA or BI plus PMA, doses of 2.5-20 μmol/L BI significantly suppressed the expression of COX-2 in a dose-dependent manner (P < 0.01). 20 μmol/L BI induced a 31% reduction in COX-2 expression in SGC-7901 cells. The concentration of PGE2 in the supernatant of SGC-7901 cells treated with PMA or PMA plus BI for 6 hours was measured. BI significantly inhibited the release of PGE2 in the supernatant of SGC-7901 cells, and a dose-response relationship was also observed. To confirm the inhibitory effect of BI on COX-2 expression, the expression of COX-2 in the mammary tumor tissues of rats fed the AIN76A diet + 9 (low), 18 (medium), and 36 (high) mmol/kg BI in a DMBA-induced breast cancer model was detected, and the results showed that the expression of COX-2 in the rat mammary tumor tissues was significantly decreased in a dose-dependent manner (P < 0.05 and P < 0.01).
