Pre-mixing antifungal agents such as zinc dimethyldithiocarbamate (Ziram) or 3-iodo-2-propynyl N-butylcarbamate (IPBC) with the culture medium when preparing nanogels usually results in better antifungal efficacy. This strategy can potentially bypass antifungal resistance and produce novel formulations with high sustained antifungal activity at similar concentrations of antifungal agents.
Encapsulation of Antifungal Agents in Carbopol Aqua SF1 Nanogels: To prepare the nanogel suspension, 0.10 g of 35 wt% stock solution is dispersed in 20 mL of deionized water, and the pH is adjusted to 8 by adding 0.25 M NaOH (aq) dropwise, monitored with a pH meter. The antifungal agent solution (75 mL of 0.2 wt% IPBC or Ziram) is gradually added to the Carbopol Aqua SF1 nanogel suspension using an injection pump while stirring continuously. The resulting aqueous suspension is stirred for an additional 30 minutes, then the pH is lowered to 5.5 by adding 0.25 M HCl (aq) solution dropwise while stirring. The suspension is centrifuged (2 x 30 minutes at 12,000 rpm). The supernatant is collected to measure the encapsulation efficiency of each antifungal agent in the nanogels, and the particles are washed twice with deionized water. The final particles are re-dispersed in deionized water using an ultrasonic bath, and the pH of the solution is adjusted to pH 8 with 1-2 drops of 0.25 M NaOH (aq). The suspension is stirred to ensure that all aggregated Carbopol nanogels swell at pH 8, then the pH is lowered to pH 5 with acetate buffer solution. The total volume of each antifungal agent encapsulated in the Carbopol Aqua SF1 nanogel suspension is maintained at a constant 100 mL.
