Exposure to 4-hydroxybenzophenone (4HBP) inhibits the viability and spheroid growth of primary mouse neural stem cells (NSCs) in vitro.
4HBP Exposure Inhibits NSC Proliferation: Primary mouse NSCs were obtained from the hippocampus of 14.5-day-old embryos and seeded into a 96-well plate containing proliferation medium. The cells were treated with a series of concentrations (EtOH, 1 nM, 10 nM, 102 nM, and 103 nM) of 4HBP for 7 days. Consistent with our previous observations, NSC spheroid formation ability was significantly impaired in the presence of 4HBP, as reflected by a marked decrease in the number of neurospheres after 7 days of culture. Additionally, NSC viability was assessed using the CCK-8 assay. 4HBP significantly inhibited the viability of mouse NSCs at concentrations within the 1 μM range. These results confirm the toxic effects of 4HBP exposure on the in vitro proliferation of NSCs.
