Ethyl vanillin (EV)/chitosan (CS)/polyvinyl alcohol (PVA) blend films were prepared by the solvent casting method. EV had a significant impact on the mechanical properties, structural properties, barrier properties, optical properties, food compatibility, and antimicrobial properties of the chitosan/polyvinyl alcohol (CS/PVA) films. Therefore, the CPEV blend has good potential to be used as a source of active films for food packaging.
Film Preparation
Ethyl vanillin (EV)-incorporated CS/PVA (CPEV) blend films were prepared by the solvent casting method. 0.5%, 1.0%, 1.5%, and 2.0% CS and PVA solutions were prepared by dissolving 0.5, 1.0, 1.5, and 2 g of CS and PVA separately in 100 ml of 1% acetic acid aqueous solution (w/v) and 100 ml of distilled water, respectively. The CS solution was kept under stirring for about 12 hours and then filtered using cheesecloth. The PVA solution was kept under stirring at 60 °C for 10 hours. A 2% EV solution was prepared by dissolving 0.2 g of EV in 10 ml of ethanol. The CS and PVA solutions were mixed in three different ratios (1:3, 1:1, and 3:1) and stirred at 60 °C for 4-5 hours until the binary blend solution became clear and homogeneous. 10 ml of the 2% EV solution was added to the resulting CS/PVA blend solution and mixed by continuous stirring until the solution became homogeneous (4-5 hours) and deaerated. The film-forming solution was poured onto Petri dishes placed on a level surface and dried at room temperature for about a week. After drying, the films were peeled off from the Petri dishes, sealed in zip-lock bags, and stored in a desiccator for further use.

Anti-angiogenic Activity
The chick embryo chorioallantoic membrane (CAM) is a commonly used in vivo model for measuring angiogenesis and anti-angiogenic activity, which was used to determine the inhibitory activity of EVA on vascular development. When 0.1, 0.3, 1.0, and 3.0 μg/egg of EVA were applied to the CAM, the inhibition rates of CAM angiogenesis were 35.3%, 46.7%, 50.0%, and 65.0%, respectively. Overall, EVA exhibited significant anti-angiogenic activity in a dose-dependent manner.
In Vivo Anti-inflammatory Activity
In the vascular permeability model, oral administration of 10, 30, and 100 mg/kg doses of EVA showed 76.0%, 79.7%, and 82.0% inhibition, respectively.
Antinociceptive Activity
The acetic acid-induced writhing response was used to detect the antinociceptive activity of EVA, in which acetic acid stimulates the pain nerve endings by releasing endogenous substances to induce pain. EVA at doses of 10, 30, and 100 mg/kg showed 26.0%, 65.8%, and 83.6% inhibition, respectively.
Inhibition of NO Generation
The inhibitory effect of EVA on LPS-induced nitric oxide (NO) production in RAW264.7 macrophages was evaluated. The accumulated nitrite in the culture medium was used as an indicator of NO levels. When the macrophages were exposed to LPS, the nitrite content increased by about 5.5-fold. Pretreatment of the macrophages with 0.1, 0.2, and 0.5 mM EVA significantly suppressed the LPS-induced NO production in a concentration-dependent manner. The inhibitory effect of EVA on NO production was also confirmed in the in vivo air pouch model.
