Preparation of DAPD-modified Magnetic Nanoparticles
Step 1. Silica-coated Fe3O4 nanoparticles
Silica-coated Fe3O4 nanoparticles were prepared by hydrolysis of TEOS on the surface of magnetic Fe3O4 nanoparticles. The precipitated Fe3O4 (2 g) obtained by co-precipitation of Fe(II) and Fe(III) was washed with anhydrous ethanol and ultrasonically dispersed in a solution containing 100 mL ethanol and 25 mL water. The pH was adjusted to 9 using ammonia water, and 4 mL of TEOS was added under vigorous stirring. After 10 h, the ferrofluid was heated to 50 °C and the hydrolysis was continued for another 12 h. A small portion was taken out to prepare a TEM sample. The precipitate was separated by an external magnetic field, the supernatant was decanted, and the product was washed with deoxygenated distilled water and anhydrous ethanol, then dispersed in 100 mL of ethanol. This sample is denoted as SCMNPs (silica-coated magnetic nanoparticles).
Step 2. DAPD-modified magnetic composite nanoparticles
The prepared composite SCMNPs were ultrasonically dispersed in a beaker. 10 mL of CPTES was added dropwise to the beaker under magnetic stirring. After hydrolysis at room temperature for 24 h, the particles were collected and washed several times with ethanol using an external magnet. The particles were redispersed in 100 mL of ethanol and transferred to a three-necked flask. Then, 2.0 g of 2,6-diaminopyridine was added, and the mixture was reacted at 50 °C for 8 h. The particles were separated using an external magnet, washed several times with ethanol and water, and dried for 2 h in a fume hood, yielding the DAPD-functionalized SCMNPs, denoted as DAPD-SCMNPs.

2,6-Diaminopyridine (0.45 g, 4.1 mmol) was dissolved in 1,4-dioxane (30 mL). 0.1 M phosphate buffer (pH 7, 10 mL) and HRP (7 mg) were added. The reaction mixture was stirred using a magnetic stirrer at room temperature, then 35% H2O2 (6 mL) was added dropwise to initiate the polymerization. After 20 hours, the precipitated product was filtered, washed with water and ethanol to remove unreacted starting materials, the enzyme, and phosphate. The product was then vacuum-dried (yield: 62%).
