SPED (Styrene-Polyethylene glycol diacrylate-9-decen-1-ol) is a novel spherical, chemically stable, highly permeable hydrophobic/hydrophilic balanced carrier for solid-phase peptide synthesis. The resin (SPED) is prepared by free-radical suspension copolymerization of the monomers styrene and 9-decen-1-ol with the amphiphilic crosslinking agent polyethylene glycol diacrylate (Mn 258).
Synthesis of SPED
Styrene is destabilized using a 1% NaOH solution to remove the inhibitor. The polymerization reaction is carried out in a four-necked reaction vessel equipped with a thermostat, a Teflon stirrer, a water condenser, and a nitrogen inlet. PVA (1.1 g) is dissolved in double-distilled water (110 mL) to prepare a 1% polyvinyl alcohol (Mw 75,000 Da) solution and added to the reaction vessel under N2 atmosphere. To prepare a 2 mol% crosslinked resin, a mixture of the monomers styrene (96 mol%, 10.99 mL), 9-decen-1-ol (2 mol%, 0.356 mL), and the crosslinking agent polyethylene glycol diacrylate (2 mol%, 0.469 mL) with toluene (8 mL) is added to the reaction vessel. Resins with different crosslinking densities are prepared; the percentage of the hydroxyl-bearing 9-decen-1-ol is maintained at 2 mol% in all crosslinking densities, and the monomers styrene and the crosslinking agent PEG diacrylate are varied. For a 4 mol% crosslinked resin, 94% styrene and 2 mol% 9-decen-1-ol are used, while for a 6 mol% resin, 92% styrene and 2 mol% 9-decen-1-ol are used, respectively. The resulting solution is stirred at a constant speed of 1200 rpm using a thermostatically controlled mechanical stirrer. Benzoyl peroxide (0.5 g) as the free-radical initiator is added to the reaction mixture and maintained under a continuous flow of N2 gas. The reaction mixture is kept at 85 °C using a thermostatic oil bath and allowed to react for 10 hours. The resulting spherical copolymer beads of 200-400 mesh size are thoroughly washed with hot water to remove PVA, Soxhlet extracted with toluene, DCM, acetone, and methanol, and vacuum-dried at 40 °C (P2O5) for 10 hours. The resin yield is 7 g.
