Coaxial nanofiber membranes loaded with astaxanthin (ASTA) have shown to promote periodontal bone defect repair while inhibiting periodontal inflammation. These membranes present a promising sustained-release drug delivery system for the treatment of periodontitis.
Preparation of the Electrospinning Solution
To prepare the electrospinning solutions, PVA powder was dissolved in distilled water at 80°C to form a 7 wt% PVA aqueous solution. Chitosan (CS) powder and varying amounts of graphene oxide (GO) dispersion were added to the PVA solution. The suspensions were stirred to achieve mass ratios of CS:PVA:GO of 10:90:0.1 and 10:90:0.5. Glacial acetic acid was then added to dissolve the CS powder, resulting in the shell-spinning solution.
Astaxanthin (ASTA) powder was dissolved in DMSO at a mass ratio of 1:10, and this solution was then mixed with the PVA solution (prepared at 80°C to a 5 wt% concentration) in a 5:90 ratio to obtain the core-spinning solution.
Both solutions were blended evenly to create the final nanofiber membrane spinning solution.
Preparation of Electrospun Nanofiber Membranes
The process for fabricating the CS/PVA/GO/ASTA coaxial electrospun nanofiber membranes is outlined in the figure. The shell and core solutions were loaded into syringes connected to a 17/22G coaxial needle. The electrospinning conditions were optimized as follows: a receiving distance of 18 cm, a voltage of 27 kV, and flow rates for the shell and core solutions of 0.2 mL/h and 0.1 mL/h, respectively.
After electrospinning for 36 hours, the nanofiber membranes collected on the collection device were dried under vacuum at 80°C for 12 hours to remove any residual solvent.
