Acetyl triethyl citrate (ATEC) is a plasticizer commonly used in pharmaceutical products and food packaging as a non-phthalate ester alternative. Due to its direct human contact and the high leakage rate from polymers, it is crucial to assess the potential health risks of ATEC at environmentally relevant low doses.
Male C57BL/6J mice were fed a diet containing ATEC at doses of 0.1 or 10 μg/kg per day for 12 weeks, to simulate realistic exposure scenarios. The study results indicated that ATEC exposure in C57BL/6J mice led to increases in body weight, body fat percentage, as well as the size of benign hepatocytes and adipocytes. Consistent with the in vivo model, ATEC treatment significantly stimulated an increase in lipid load within mouse and human hepatocytes. Mechanistically, ATEC induced the transcriptional expression of genes involved in de novo lipogenesis and lipid absorption.

Acetyl triethyl citrate (ATEC) is a water-soluble plasticizer used in pharmaceutical plastic polymers. The pharmacokinetics and metabolism of ATEC in rats were studied using liquid chromatography-tandem mass spectrometry (LC-MS/MS).
After intravenous administration of 10 mg/kg and oral administration of 500 mg/kg doses of ATEC to rats, the concentration of ATEC in plasma rapidly declined and became undetectable after 0.5 hours and 6 hours, respectively. The volume of distribution (Vz) was 4.8 L/kg, which is higher than the volume of body water, suggesting possible distribution into extravascular spaces. The absolute bioavailability (F) of ATEC was 14.8%. This relatively low F value indicates poor absorption of ATEC in the gastrointestinal tract, degradation within the intestine, and significant first-pass effects.
After intravenous administration, ATEC was rapidly eliminated, with an apparent terminal half-life of 0.03 hours and a mean residence time of 0.03 hours, while for oral ATEC, the apparent terminal half-life was 1.03 hours and the mean residence time was 6.56 hours.
