The COVID-19 pandemic has significantly impacted global healthcare, leading to shortages of essential drugs, including propofol. In response, various protocols for propofol synthesis have been proposed in recent years. In this study, we present a novel process that utilizes paracetamol (also known as acetaminophen), a widely available and commonly used active pharmaceutical ingredient, as the starting material. By performing the initial three steps-Friedel-Crafts double alkylation, acetyl deprotection, and diazotization-in acidic media, we developed an efficient one-pot synthesis approach for propofol.
