Tolnaftate (Tf), an antifungal drug, was combined with graphene nanoplatelets (Gn) to create a composite material (Tf-Gn) using a wet chemical method. This composite was then incorporated into a biocompatible polymeric blend of Eudragit RL100/Eudragit RS100 (EuRL100/EuRS100) and processed by electrospinning to produce nonwoven nanoengineered scaffolds (nanofibers) with enhanced anti-dermatophytic properties.
Procedure:
Tolnaftate (Tf) was efficiently loaded onto graphene nanoplatelets (Gn) through a wet chemical route to form the Tf-Gn composite. Briefly, a 1 mg/mL solution of Tf was added to a uniform dispersion of Gn in acetone and subjected to sonication for a few seconds. The resulting dispersion was stirred overnight at room temperature in the dark and then ultracentrifuged at 15,000 rpm for 30 minutes. The loading capacity of Tf was determined by measuring the concentration of unbound Tf in the supernatant using a UV-visible spectrophotometer (Shimadzu, 1700), based on the linear equation (y = 0.0606 + 0.0075).
