Guaiac resin, derived from Guaiacum officinale L. and G. sanctum L., has long been valued in traditional medicine, yet its bioactive constituents remain underexplored. Recent research focused on isolating and characterizing lignans from guaiac resin, with particular attention to their anti-inflammatory potential. Guided by nitric oxide (NO) suppression assays in interleukin-1β (IL-1β)-stimulated rat hepatocytes, six lignans were successfully purified: dehydroguaiaretic acid, (±)-trans-1,2-dihydrodehydroguaiaretic acid, furoguaiaoxidin, meso-dihydroguaiaretic acid, furoguaiacin, and nectandrin B.
This study represents a milestone in phytochemical exploration. Dehydroguaiaretic acid was reported for the first time as a non-derivative constituent of guaiac resin, while furoguaiaoxidin was newly isolated from a natural source. Additionally, nectandrin B and trans-1,2-dihydrodehydroguaiaretic acid were first identified in guaiac resin. All six lignans displayed significant NO-suppressing activity, suggesting that they underpin the resin's anti-inflammatory properties. Since NO suppression is a standard marker for evaluating anti-inflammatory efficacy, these findings provide strong mechanistic evidence for the pharmacological relevance of guaiac resin.
The isolation procedure combined silica gel column chromatography, Sephadex LH-20 separation, preparative thin-layer chromatography, and preparative HPLC, underscoring the resin's chemical complexity. Collectively, these results highlight guaiac resin as a promising source of bioactive lignans with potential therapeutic applications in inflammatory disorders, offering new opportunities for both natural product chemistry and medicinal research.
